What is peptide receptor radioligand therapy?

Peptide receptor radioligand therapy (PRRT) is a type of molecular targeted radiation therapy used to treat certain types of cancer. It involves administering a radioactive substance called a radioligand, which binds to specific receptors on cancer cells and delivers targeted radiation to kill those cells.
The process begins by identifying cancers that have an abundance of certain receptors on their cell surfaces, such as somatostatin receptors or glucagon-like peptide-1 (GLP-1) receptors. These receptors can be detected using molecular imaging techniques like PET scans. Radioligands are then developed that specifically bind to those receptors.

These radioligands are chemically linked to a radioactive isotope, usually Lutetium-177 or Yttrium-90. When injected into the patient's bloodstream, the radioligands circulate and selectively bind to receptors on cancer cells. The localized radiation kills cancer cells by damaging their DNA. The radioactivity breaks chemical bonds within cancer cell molecules, interfering with critical cell functions and causing cell death.
